Sodium starch glycolate is a white or nearly white, free-flowing, and highly hygroscopic powder. According to PhEur 6.0, microscopic examination shows irregular, ovoid, pear-shaped, or rounded granules ranging from 10–100 μm in size. Occasionally, compound granules containing 2–4 components may also be observed. The granules feature an eccentric hilum with clearly visible concentric striations. Under crossed Nicol prisms, they display a characteristic black cross intersecting at the hilum, while small crystals can be seen on the granule surfaces. The granules exhibit remarkable swelling behavior when exposed to water.

Sodium starch glycolate is widely utilized in oral pharmaceutical formulations as a disintegrant for tablets and capsules. It is suitable for tablets produced by both direct compression and wet granulation methods. The typical usage level ranges from 2% to 8%, with an optimal concentration of around 4%, although in many formulations 2% is often sufficient. Its disintegration mechanism is based on rapid water absorption followed by strong and rapid swelling.
Unlike many other disintegrants whose performance may be reduced by hydrophobic excipients such as lubricants, sodium starch glycolate maintains its disintegration efficiency. In addition, increased tablet compression force has little to no impact on disintegration time.
It has also been studied for potential application as a suspending agent.
